Cockayne syndrome (CS) is a recessive progeroid syndrome stemming from non-functional mutations in the chromatin remodeller protein, CSB. CSB is critical for recognising DNA damage stalled transcription, to coordinate RNA polymerase II (Pol II) processing and recruit downstream DNA repair factors. My work aims to elucidate the unknown molecular events in CSB-mediated DNA repair by developing an in vitro transcription system utilising novel fluorescent RNA aptamers to monitor transcription restart. Additionally, I am using biochemical assays to dissect the biochemical activities of CSB domains with the aim to perform a fragment-based screen to explore therapeutic inhibition.
